老化細胞では、あえて細胞質に核由来DNAを蓄積させている? (Nature Communications 2018年4月28日号掲載論文)

結論から言うと、老化細胞において、(p16の下流で抑制されているE2F転写因子のターゲットである)DNaseの発現が低下することで、細胞質にDNAが蓄積し、cGAS-STING経路を介してSASPを誘導されていることを示した論文。

 

少し学会に行っていたので間があきました(論文読んでるストックがあるので間を埋め合わせるつもりです。日本人著者の論文が増えるのはご愛嬌)。

 

本日は「Downregulation of cytoplasmic DNases is implicated in cytoplasmic DNA accumulation and SASP in senescent cells (細胞質DNaseのダウンレギュレーションによる細胞質DNA蓄積と老化細胞におけるSASPとの関係)」という論文で、大阪大学微生物病研究所 環境応答研究部門 遺伝子生物学分野 原英二先生のグループによる研究。(論文サイトへのlink→*1

個人的には、DNaseの発現制御が、ちゃんと老化マーカーのp16の下流の転写因子とつなげられていたので、読んでいてスッキリしました。p16がpositiveな老化細胞なら、下流でDNaseが抑制されている可能性が高いと言えるかも(もちろん他の転写因子などによっても制御され得るので断言はできません)。

あと、老化細胞においては、(経時的な)核膜の損傷によってゲノムDNAが漏洩しやすくなっていたりするのだろうか?ぱっと調べても出てこなかったので有識者の方教えてください。

Accumulating evidence indicates that the senescence-associated secretory phenotype (SASP) contributes to many aspects of physiology and disease. Thus, controlling the SASP will have tremendous impacts on our health. However, our understanding of SASP regulation is far from complete. Here, we show that cytoplasmic accumulation of nuclear DNA plays key roles in the onset of SASP. Although both DNase2 and TREX1 rapidly remove the cytoplasmic DNA fragments emanating from the nucleus in pre-senescent cells, the expression of these DNases is downregulated in senescent cells, resulting in the cytoplasmic accumulation of nuclear DNA. This causes the aberrant activation of cGAS-STING cytoplasmic DNA sensors, provoking SASP through induction of interferon-β. Notably, the blockage of this pathway prevents SASP in senescent hepatic stellate cells, accompanied by a decline of obesity-associated hepatocellular carcinoma development in mice. These findings provide valuable new insights into the roles and mechanisms of SASP and possibilities for their control.

(私訳と勝手な注釈) 

老化に関連した分泌表現型(SASP)が生理学と病気の多くの側面に寄与するという知見が蓄積してきています。したがって、SASPの制御は、私たちの健康に多大な影響を及ぼします。一方、SASPの制御の理解は完全ではありません。本論文では、核DNAの細胞質蓄積がSASPの発症に重要な役割を果たすことを示します。 DNase2とTREX1はどちらも、pre-senescent cells(老化前細胞?)の核から発生する細胞質DNA断片を迅速に除去するDNaseであるが、これらの発現は老化細胞でダウンレギュレートされ、核DNAの細胞質蓄積をもたらします。核DNAの蓄積は、cGAS-STING細胞質DNAセンサーの異常な活性化を引き起こし、インターフェロン-βの誘導を通じてSASPを誘発します。特に、この経路を阻害すると、マウスの肥満関連肝細胞癌の発達の低下を伴う老化肝星細胞のSASPが阻害されます。これらの発見は、SASPの役割とメカニズム、およびその制御の可能性に新しい知見を提供します。
